I have read with interest the discussion so far, and would like to add some personal views based on our work and reading the literature.
I critical issue of metal coordination bonds in the weak M-L bonds, compared to covalent bonds. However, this is a thermodynamic phenomenon.
For a number of metal ions, the kinetics dominate the reactivity, and in fact Pt(II0, Pt(IV), and also Ru(II) and Ru(III) do have ligand exchange kinetics that are in the same range as biological processes.
Other metals are easily 1000 times slower (as Iridium), 1000 to 1000000 times faster, such as Pd and Ni.
For metal complexes to treat diseases, such as cancer, I feel that lots of exciting possibilities, including targeting drugs, will become available, as soon as we have better control of the kinetics in biological systems.
